The Off-Broadway Production of Hamilton was the first production of the musical as it is, with the current characters and songs; the first production, The Hamilton Mixtape, including many songs that were cut for the Off-Broadway show.
Hamilton during Off-Broadway was located at The Public Theater, and later resigned and made its debut on Broadway, including the same songs (albeit with minor lyrical and musical changes), characters, and cast.
